Abstract

Derivation of autologous stem cells has become an area of intense interest, due to their potential application in human cell therapy and reproductive technologies in various animal species. Kues et al. (2005) have recently reported the isolation of fetal somatic stem cells from murine and porcine tissues. These cells have been shown to form 3 dimensional colonies under special growth conditions as well as the expression of stem cell specific markers. Using similar culture techniques, we were able to isolate putative stem cells from adult bovine skin biopsies. These cells also exhibit the formation of 3 dimensional colonies, and clonal growth. Furthermore, expression of pluripotency markers Oct-4 and SSEA-1 have been confirmed by RT-PCR using bovine specific primers. Alkaline Phosphatase expression has been confirmed by staining. Characterization of the differentiation potential of these cells is needed to determine the suitability of this population for use in autologous stem cell therapies and animal reproductive technologies.
