Abstract

The blooming of advanced kinase inhibitors in the past few years has urged more efforts for novel, selective drugs with potential activity in different pathological conditions. Mitogen activated protein kinases (MAPKs) are very important targets for different diseases. p38 is one of MAPKs. Since its discovery in 1994, the pivotal role of p38 in production of proinflammatory cytokines has attracted many researchers to produce a variety of p38 inhibitors with different chemical structures and binding fashions to p38. These inhibitors are mostly indicated for inflammatory diseases, such as rheumatoid arthritis. However, the role of p38 in cancer has been investigated lately. This review discusses different chemical structures of p38 inhibitors and their structure activity relationships. Also, binding fashions of these molecules to p38 are illustrated. Moreover, the role of p38 inhibitors in apoptosis and incancer treatment is presented.
