Abstract

Eastern Russell’s viper (Daboia siamensis) venom of Myanmar and Indonesian origin were separated by cation exchange chromatography, and further resolved by SDS-PAGE. Relevant bands were excised and digested with trypsin digestion and analysed by tandem mass spectrometry (MS/MS). Identified fragments were matched to known Russell’s viper proteins within the UniProt database. Venom in human plasma was purified via cation exchange and size exclusion chromatography and SDS-PAGE. This is the first study to explore a novel proteomic method to isolate and detect D. siamensis venom proteins and peptides in human plasma. It is the first comparative proteomic analysis of Myanmese and Indonesian D. siamensis venom, and the results provide a framework to elucidate the underlying effects of Eastern Russell’s viper envenoming in victims of different geographical origins.
