Abstract
Teratoma formation is an ideal in vivo model for evaluating the differential potential of cells, with pluripotent cells giving rise to derivatives of all three germ layers i.e. endoderm, mesoderm and ectoderm. In this report, we compared in vivo differentiation potential of 2N and 4N ESCs. Embryonic stem cells i.e. 2N and 4N, were injected into the testis capsule of anaesthetized severe combined immuno-deficient (SCID) mice ( 0.5 3 106 cells per testis). At 5.5 weeks post-injection, the animals were humanely killed and teratomas were excised. The teratoma explants were either plated in culture dishes in simple MEF medium and allowed to expand to assess their respective ploidy, or were fixed and embedded in paraffin blocks for morphological and immuno-histochemical analyses. Morphological and immuno-histochemical analyses of teratomas confirmed presence of cells indicative of all three germ layers in both types of teratomas. Further, the 2N and 4N teratoma derived cell maintained the respective ploidy of the injected pluripotent cells. In conclusion, this study has demonstrated that the 4N ESC can form teratomas in a manner similar to 2N ESC and can give rise to derivatives of all 3 germ layers.
