Abstract

Self-expanding stents are commonly used to relieve gastrointestinal (GI) obstructions caused by tumours. However, continued tumour ingrowth can result in stent blockage and failure. To address this problem, we developed novel drug-eluting stents (DESs) that simultaneously provide support and the controlled release of anticancer drugs over a prolonged period to prevent ingrowth and reduce tumour load. A simple and versatile coating technique was developed that could be applied to various types of GI stents and allowed multiple coating formations to be applied in sequence, providing DESs with high drug loadings and long release periods (up to 160 days). In vitro studies revealed that the drugs released from the DESs were highly cytotoxic to human colon cancer cells. These promising results indicate that the DESs could potentially reduce tumour burden and improve patients’ quality of life.
