Abstract
This thesis describes a novel and adaptable strategy that can be applied to render non-responsive micelles pH-responsive, resulting in abrupt cargo release at specific pH values compatible with endosomal delivery. Oligoelectrolytes and drugs were co-encapsulated in micelles with high encapsulation efficiencies and following a decrease in the pH and oligoelectrolyte ionization, both were ejected from the micelle core. The oligoelectrolyte-mediated, pH-triggered release of drugs from the micelles resulted in significant increases in cytotoxicity to cancer cell lines, which may also be facilitated by accelerated endosomal escape of the drugs caused by disruption of the endosomal membranes. Furthermore, we show how this approach is broadly applicable to non-responsive micelles regardless of their composition, and various classes of hydrophobic chemotherapeutics.
