Abstract
Antimicrobial resistance is one of the biggest threats to modern medicine and public health according to global organizations such as the World Health Organization. Hence new approaches to combat drug resistant bacteria and address the shortage of new antimicrobials are desperately needed, particularly for Gram-negative bacteria. This research investigated the identification and evaluation of synthetic and repurposed compounds as inhibitors of important resistance mechanisms: the RND efflux pumps in Gram-negative bacteria including the AcrAB-TolC efflux pump in E. coli and MexAB-OprM efflux pump in Pseudomonas. As a result, six compounds among thirty-nine benzo[h]chromene and flavonoid derivatives were identified as putative inhibitors for AcrB. IvacaPor, a transmembrane conductance protein potentiator, was identified as a promising candidate for repurposing as inhibitor for MexB.
