Abstract

Methotrexate (MTX) is most notably used for the treatment of the paediatric cancer acute lymphoblastic leukemia (ALL), breast cancer and osteosarcoma. Despite a high rate of success in the cancer treatment, long-term adverse effects of chemotherapy on bone/bone marrow such as bone loss, marrow adiposity and bone marrow vasculature injuries have been reported among cancer patients and in experimental studies. However, the underlying molecular mechanisms of damage and recovery potential are poorly understood. Current research with in vitro and in vivo studies and using a vast range of techniques has identified that deregulation of Notch2 signalling plays an important role in mediating MTX-induced bone/bone marrow damage. Our findings illustrate that targeting Notch2 could be a potential treatment option to overcome MTX chemotherapy bone-related adverse effects.
